Naturforsch. 40c, 705—709 (1985); received May 29, 1985 Chloroplast, Thylakoid Membrane Proteins, Electrophoresis, Polyacrylamide, Coomassie Blue Stain 14C labeled thylakoid membranes of Viciafaba chloroplasts were separated by one-dimensional electrophoresis in gel cuvettes of different length using a 12 — 18% polyacrylamide gradient gel system containing 0.1% sodiumodecylsulfate and 6 m urea. The use of gels with a length of 22 cm and 60 cm led to the detection of about 40 and 80 polypeptide bands, respectively, after dye staining and 50 and more than 100 polypeptide bands, respectively, after autoradiography. The conditions used strongly denaturate chlorophyll proteins and avoid the formation of different polypeptide conformations. Polypeptides differing in were separated in sharp bands.

Using a Temperature Gradient against the Time in Polyacrylamide Gel Electrophoresis May Eliminate the Need for Stacking Gels
Background and Objectives: Making stacking gels for polyacrylamide gels in the laboratory by conventional methods is laborious and time consuming. Considering the role of temperature in polyacrylamide gels with respect to electrical resistance and viscosity, we assumed that decreasing the temperature would cause an increase in electrical resistance and viscosity. Ultimately, a downward tempera...
متن کاملSynthesis of thylakoid membrane proteins by chloroplasts isolated from spinach. Cytochrome b559 and P700-chlorophyll a-protein
Intact chloroplasts, purified from spinach leaves by sedimentation in density gradients of colloidal silica, incorporate labeled amino acids into at least 16 different polypeptides of the thylakoid membranes, using light as the only source of energy. The thylakoid products of chloroplast translation were visualized by subjecting membranes purified from chloroplasts labeled with [35S]methionine ...

متن کاملChanges in Thylakoid Galactolipids and Proteins during Iron Nutrition-Mediated Chloroplast Development.
Changes in the amounts of thylakoid galactolipids and proteins were monitored for 96 hours following iron resupply to iron-deficient sugar beet (Beta vulgaris L. cv F58-554H1) plants. During this period of iron nutrition-mediated chloroplast development, the amount of galactolipid per leaf area increased linearly with time. Assuming galactolipids are an index for the amount of thylakoids, then ...
متن کاملThe sites of synthesis of the principal thylakoid membrane polypeptides in Chlamydomonas reinhardtii
The sites of synthesis of the major thylakoid membrane polypeptides have been studied in the green alga Chlamydomonas reinhardtii by pulse labeling of cells with [14C]acetate in the presence of inhibitors specific for chloroplast and cytoplasmic protein synthesis. The labeled membrane polypeptides were separated by an improved method of sodium dodecyl sulfate (SDS) gradient gel electrophoresis,...
